What quantity of antigen is required for antigen affinity purification?

For purification of 50 ml antiserum, about 2-10 mg of protein or 3-10 mg of lyophilized peptide are required to couple to the affinity matrix column. The yield of antibody produced can range from 5 to 10 % of total immunoglobulins and is based on quality of immune response. Generally, a minimum of 10 mg of antibody can be obtained.
